    Directions:

    Suspend 37.4g of the medium in one liter of distilled water or deionized water. Mix well and heat with frequent agitation. Sterilize in an autoclave at 121℃(15 lbs.) for 15 minutes.

    Principle and Interpretation:

    Eosin-Y and methylene blue makes the medium slightly selective and inhibit certain gram-positive bacteria. Both dyes act as indicator and inhibiting agent. These dyes differentiate between lactose fermenters and non-fermenters. Eosin Y and methylene blue forms a complex at acidic pH, which acts as inhibiting agent. Essential nutrients and growth factors are provided by pancreatic digest of gelatin. Phosphates acts as good buffering agent. E.coli forms colonies with green metallic sheen, indicating strong lactose fermentation.

    Appearance:

    Dehydrated medium is a free-flowing purplish red powder. The prepared medium is a kind of purplish red brown gel.

    Precautions:

    This medium is for laboratory use only. Dried medium which is past shelf life, caking or color variation cannot be used.

    Storage conditions and Shelf life:

    Levine EMB Agar must be stored tightly capped in the original container at 5-30°C. The dehydrated medium has a shelf life of 3 years from date of manufacturing. Prepared medium may be stored, out of direct light at 2-8°C.

    Quality control:

    Inoculate and incubate at 30-35℃for 24-48 hours.
